Real time observation and simulation of conformational changes of E6AP HECT domain.

The HECT domain repeats spherical and elliptical conformations through the flexible region. Computer simulation results show that the spherical shape corresponds to the catalytic structure and the oval shape corresponds to the L-shaped structure. The formation efficiency of the ubiquitin dimer (Ub2) changes significantly depending on the conformational state of the flexible hinge region.
