Prime editing is used to treat the genetic disease with specific pathogenic mutation. Some websites may help to design a variety of candidate pegRNAs and nsgRNAs, which are then constructed by means of the golden gate assembly technique. Subsequently, these plasmids are applied for in vitro screening to choose the pegRNA and nsgRNA with best performance. These well-performed RNAs are then constructed into dual-AAV system. After injected into the mice model, the editing efficiency and off-target events of prime editing are finally be evaluated.
