Through the precise molecular design of cationic polypeptides, we have prepared a new dynamic nanomedicine (or nanomachine) that loosely encapsulates a single molecule of oligonucleotide drug. When administered intradermally, the nanomedicine can efficiently deliver the oligonucleotide payload to tumor-draining (sentinel) lymph nodes, enhancing cancer immunotherapy through activation of immune cells.
