How gene dependency analysis reveals therapeutic vulnerabilities in breast cancer. Part I integrates a multi-omics cohort of breast cancer cell lines and establishes a dependency-marker association analysis pipeline. Part II deconvolutes statistically derived DMAs from dual biologically interpretable perspectives to pinpoint therapeutically relevant targets. Part III further delineates two core dependency-based clusters, characterizes molecular mechanisms underlying cluster-specific dependency signatures, and identifies DMAs within each cluster, to inform precision therapeutic strategies. This study highlights how DMA analysis can help elucidate fundamental cell-biological mechanisms and uncover clinically actionable therapeutic targets.
