The key components of the dual-mode CapCell are shown in Fig. A. Fig. B and C show the simulated and experimental widefield and high-resolution images with the optimized illumination geometry, which resulted in a 12-fold increase in optical power compared to widefield imaging, due to the concentration of the light within the targeted region of interest (ROI). The resolution and FOV of the CapCell was determined using a USAF resolution target as shown in Fig. D. The widefield mode achieved a resolution of 13 μm and an FOV of 11.2 mm × 6.3 mm, while the high-resolution mode achieved a resolution of 5.5 μm and an FOV of 2.8 mm × 1.5 mm. To demonstrate the ability to resolve vascular structures, we used green light illumination to image 4T1 tumor-bearing mice implanted with mammary window chambers with widefield and high-resolution settings Fig. E
