RNA-based genetic switches exhibit leaky expression even in the 'OFF' state, resulting in narrow dynamic range and genetic instability (top). SUPER adds synthetic sRNA as an auxiliary module to suppress leaky expression, and by modulating sRNA expression levels, enables various operating modes including complete repression, leakage minimization, and performance enhancement (bottom).
