Caption
RUNX1 turns POSTN+ fibroblasts into stress-resistant scar builders. In periostin-positive (POSTN+) fibroblasts, RUNX family transcription factor 1 (RUNX1) transcriptionally activates activating transcription factor 4 (ATF4), periostin (POSTN), and midkine (MDK). ATF4 reinforces endoplasmic reticulum (ER) stress adaptation; POSTN sustains pathogenic fibroblast activation; MDK promotes angiogenesis. The solute carrier family 7 member 11 (SLC7A11)/glutathione peroxidase 4 (GPX4) axis reduces lipid reactive oxygen species (ROS), supporting ferroptosis resistance and linking stress survival to matrix remodeling and vascular niche changes in skin fibrosis.
