Structure-guided rational design of the common-light-chain EGFR×c-MET bispecific antibody ALK101 (IMAGE)
Caption
Starting from the parental antibodies necitumumab (anti-EGFR) and MM-131 (anti-c-MET), researchers identified incompatible light-chain residues, rebalanced dual-antigen binding through targeted back-mutations and saturation mutagenesis, and restored EGFR affinity via heavy-chain CDRH3 maturation — yielding the common-light-chain bispecific antibody ALK101 without large-scale library screening.
