Figure 1 | Dual-view Bessel two-photon projection microscopy (dv-B2PM) and three-dimensional neuronal reconstruction. a, two objectives arranged at 90 degrees collect orthogonal projections of the same three-dimensional volume. OBJ1 delivers the Bessel two-photon excitation beam and collects fluorescence for photomultiplier-tube detection, while OBJ2 collects the second projection and delivers the two-photon uncaging beam. b, conventional light-sheet imaging forms a volume from sequential optical sections. c, dv-B2PM records two synchronized projections of the full volume in a single volume scan. d, calcium activity projections recorded by the camera and photomultiplier tube (PMT) after glutamate uncaging. e, major neuronal structures, including the soma and dendritic branches, traced in the two projection views. f, orthogonal projections enable reconstruction of the neuron's 3D geometry.
