The Tsc1/2 complex inactivates Rheb. A mutation in Tsc2 results in Rheb activation, inducing neuron morphological defects and dysfunctions. These abnormalities promote memory impairment in TSC mice. Lonafarnib inhibits the farnesylation of Rheb, which is necessary for its activation, suppressing Rheb activation and thereby normalizing nerve morphology, function, and memory in TSC mice.
