(Top) Action mechanism of Tailon. Tailon sequences in mRNA bind to TENT4 or PAP proteins. These enzymes extend the poly(A) tail, helping stabilize the mRNA. (Bottom left) The amount of RNA remaining in cells was measured over time. Control linear mRNA was rapidly degraded, whereas Tailon-containing linear mRNA remained at levels comparable to circular RNA. (Bottom right) Total protein production was measured for linear mRNAs and circular RNA. Linear mRNA containing a tailon produced five times more protein than conventional linear mRNA and approximately nine times more than circular RNA.
