Semaglutide, the active ingredient in GLP-1 weight loss drugs, directly improves liver function through a specific subset of liver cells called liver sinusoidal endothelial cells (LSECs), independently of any weight loss. Contrary to the long-held assumption that liver cells lack the receptor semaglutide binds to, the study found that LSECs do carry this receptor, and when activated, they release anti-inflammatory molecules that calm the broader liver environment, reducing inflammation and scarring associated with metabolic liver disease (MASH). The findings suggest that physicians may be able to prescribe lower doses of GLP-1 medicines to treat liver disease — avoiding the side effects and costs of higher doses needed for significant weight loss — and that liver health, not just weight, should be a key measure of treatment success.

Acute myeloid leukemia (AML) has a poor prognosis, especially in elderly and relapsed/refractory patients, with 5‑year survival below 30%. Chemotherapy alone suffers from off‑target toxicity and drug resistance. Immunotherapy offers precision and durable effects but shows limited efficacy as monotherapy. Combining chemotherapy with immunotherapy is emerging as a promising paradigm. This review summarizes the rationale and advances of combining immune checkpoint inhibitors, CAR‑T, CAR‑NK, CAR‑macrophage therapies, antibody‑drug conjugates, bispecific antibodies, and cancer vaccines with chemotherapy for AML. We discuss preclinical and clinical progress, core challenges (off‑target toxicity, tumor heterogeneity, variable efficacy), and future directions toward precision medicine.
